Identifying population differences in diffusion MRI structural networks
 
Amyotrophic lateral sclerosis (ALS), the most common form of motor neurone disease, is a devastating neurodegenerative disorder. Network analyses of the connectivity between brain regions may be useful in understanding the white matter degeneration associated with the disease. We investigated the connectivity between brain regions in a group of 30 ALS patients when compared with a group of age-matched healthy controls. 85 grey matter regions (network nodes) were identified from structural magnetic resonance imaging (MRI) and network connections were formed from the white matter tracts generated by diffusion MRI and probabilistic tractography. A non-parametric technique, called network-based statistics, identified a 13 node subnetwork of reduced connectivity in the ALS group when compared with the controls involving connections to motor, prefrontal and subcortical regions.
