The regulation of protein synthesis is a critical determinant of the cellular proteome and how it is altered in response to extracellular stimuli. The control of the elongation phase of protein synthesis, by the Ca2+/calmodulin dependent elongation factor 2 kinase (eEF2k), is important for resistance to stresses such as ischemia (Romero-Ruiz et al., 2012) or nutrient deprivation (Leprivier et al., 2013), and in memory formation (Gildish et al., 2012). Whilst previous work has identified various upstream regulators of eEF2k (Proud, 2007), little is known about the dynamics of its control or how the various cell signalling inputs are integrated to modulate eEF2k activity. We used kinetic modelling of the known interactions between the MEK/ERK, mTORC1 and eEF2k pathways and found that such a model successfully fit the experimental data. Finally, we tested the model by performing an additional experiment in which we inhibited the primary phosphatase in the model and found that the model could fit the empirical data.
