"Transcriptome-wide modelling of RNA life cycle"
Cells regulate gene expression through a complex coordination of processes. Understanding the relative balance and dynamical control of production and decay of macromolecules is central for dissecting its mechanisms. Partly due to the lack of direct measurements underlying degradation, most model-based efforts focus solely on changes in transcription and assume a constant degradation rate. We used χCRAC , an improved experimental protocol, to collect rapid time series of RNA binding to degradation factors under stress. We discovered that a large fraction of the transcriptome displays dynamic binding behaviour, calling into question earlier modelling assumptions. We are now using these and other relevant data to develop a transcriptome-wide state-space model of RNA life cycle with time-variable transcription and degradation rates.
